Issue facing in programming stm32f469bit series microcontroller custom pcb. One time programming is fine , later it cannot be programmed. why is this isuue?

You got to be a bit more specific here. What does not work, what tools you use, and what error messages are displayed ?

Perhaps your code hardfaults immediately, or you have RDP (readout protection) enabled.
